ScienceDaily (Mar. 29, 2012) ? How do you build a brain? In the March 30 issue of Science a team of investigators presents a surprising answer, reporting their discovery of a remarkably simple organizational structure in the brains of humans and other primates. Employing sophisticated mathematical analysis of advanced imaging data, they found that the pathways carrying neural signals through the brain are arranged not in a disorganized tangle but in a curved, three-dimensional grid.

"We found the brain is built from parallel and perpendicular fibers that cross each other in an orderly fashion. Finding this kind of simple organization in the forebrain of higher animals was completely unsuspected," says Van Wedeen, MD, of the Martinos Center for Biomedical Imaging at Massachusetts General Hospital, who led the study. "Knowing there is a simple plan that, modified by evolution and development, gives rise to all brains has implications for researchers working to build an atlas of brain connections, for pursuing investigation of how the brain develops and for expanding theories of how the brain works."

It is well known that neural pathways in the spinal cord and brain stem are organized in three principal directions -- head to tail, side to side, and front to back -- all of which are either parallel or perpendicular to each other and reflect the basic patterns of embryonic development. But following those pathways into the brains of higher animals -- particularly into the cerebral cortex -- has been challenging because each pathway crosses many others within a small space. Previous studies that followed the movement of tracer chemicals injected into specific brain regions could track the progress of single pathways only and could not be used in human participants.

In the current study, Wedeen and his colleagues used diffusion spectrum MR imaging -- a technology he developed that reveals the orientation of all fibers that cross a particular point on a pathway -- coupled with mathematical analysis of all crossing or adjacent pathways in the brains of four species of non-human primates and in human volunteers. Wedeen's previous studies of animal brains had revealed parallel crossing pathways that appeared to form sheet-like structures, but it was not clear whether those structures were pervasive or only characterized a few brain systems.

The analysis revealed that all crossing or adjacent fibers were either perpendicular or parallel to the original pathway. Each of the crossing fibers was, in turn, crossed by its own perpendicular fibers, interwoven like the threads in a sheet of fabric, that defined box-like, three-dimensional curved grid structures. The same grid-like pattern was seen throughout the white matter of the brains of all four studied non-human primates -- rhesus monkeys, owl monkeys, marmosets and galagos -- and the human volunteers.

"I don't think anyone suspected the brain would have this sort of pervasive geometric pattern," Wedeen says. "Although our findings could be described as a new longitude and latitude for the brain, they're also leading us to an entirely new understanding of how and why the brain is organized the way it is. The old image of the brain as a tangle of thousands of discrete, unconnected wires didn't make sense from an evolutionary standpoint. How could natural selection guide each of those wires into more efficient, advantageous configurations?

"The very simplicity of this grid structure is the reason why it can accomodate the random, gradual changes of evolution," he continues. "It's easier for a simple structure to change and adapt, whether we're talking about the big changes that occur across evolution or the changes that can occur during an individual's lifetime -- both the normal neuroplasticity associated with development and learning or the damage that results from injury or disease. A simple grid structure makes both evolutionary and develomental sense." Wedeen is an associate professor of Radiology at Harvard Medical School and on the faculty of the Harvard-MIT Health Sciences and Technology Program.

DENVER (AP) ? Angeline Chilton says she can't drive unless she smokes pot. The suburban Denver woman says she'd never get behind the wheel right after smoking, but she does use medical marijuana twice a day to ease tremors caused by multiple sclerosis that previously left her homebound.

"I don't drink and drive, and I don't smoke and drive," she said. "But my body is completely saturated with THC."

Her case underscores a problem that no one's sure how to solve: How do you tell if someone is too stoned to drive?

States that allow medical marijuana have grappled with determining impairment levels for years. And voters in Colorado and Washington state will decide this fall whether to legalize the drug for recreational use, bringing a new urgency to the issue.

A Denver marijuana advocate says officials are scrambling for limits in part because more drivers acknowledge using the drug.

"The explosion of medical marijuana patients has led to a lot of drivers sticking the (marijuana) card in law enforcement's face, saying, 'You can't do anything to me, I'm legal,'" said Sean McAllister, a lawyer who defends people charged with driving under the influence of marijuana.

It's not that simple. Driving while impaired by any drug is illegal in all states.

But it highlights the challenges law enforcement officers face using old tools to try to fix a new problem. Most convictions for drugged driving now are based on police observations, followed later by a blood test.

Authorities envision a legal threshold for pot that would be comparable to the blood-alcohol standard used to determine drunken driving.

But unlike alcohol, marijuana stays in the blood long after the high wears off a few hours after use, and there is no quick test to determine someone's level of impairment ? not that scientists haven't been working on it.

Dr. Marilyn Huestis of the National Institute on Drug Abuse, a government research lab, says that soon there will be a saliva test to detect recent marijuana use.

But government officials say that doesn't address the question of impairment.

"I'll be dead ? and so will lots of other people ? from old age, before we know the impairment levels" for marijuana and other drugs, said White House drug czar Gil Kerlikowske.

Authorities recognize the need for a solution. Marijuana causes dizziness, slowed reaction time and drivers are more likely to drift and swerve while they're high.

Dr. Bob DuPont, president of the Institute for Behavior and Health, a non-government institute that works to reduce drug abuse, says research proves "the terrible carnage out there on the roads caused by marijuana."

One recent review of several studies of pot smoking and car accidents suggested that driving after smoking marijuana might almost double the risk of being in a serious or fatal crash.

And a recent nationwide census of fatal traffic accidents showed that while deadly crashes have declined in recent years, the percentage of mortally wounded drivers who later tested positive for drugs rose 18 percent between 2005 and 2011.

DuPont, drug czar for Presidents Richard Nixon and Gerald Ford, wrote a paper last year on drugged driving for the Obama administration, which has made the issue a priority.

Physicians say that while many tests can show whether someone has recently used pot, it's more difficult to pinpoint impairment at any certain time.

Urine and blood tests are better at showing whether someone used the drug in the past ? which is why employers and probation officers use them. But determining current impairment is far trickier.

"There's no sure answer to that question," said Dr. Guohua Li, a Columbia University researcher who reviewed marijuana use and motor vehicle crashes last year.

His survey linked pot use to crash risk, but pointed out wide research gaps. Scientists do not have conclusive data to link marijuana dosing to accident likelihood; whether it matters if the drug is smoked or eaten; or how pot interacts with other drugs.

The limited data has prompted a furious debate.

Proposed solutions include setting limits on the amount of the main psychoactive chemical in marijuana, THC, that drivers can have in their blood. But THC limits to determine impairment are not widely agreed upon.

Two states place the standard at 2 nanograms per milliliter of blood. Others have zero tolerance policies. And Colorado and Washington state are debating a threshold of 5 nanograms.

Such an attempt failed the Colorado Legislature last year, amid opposition from Republicans and Democrats. State officials then set up a task force to settle the question ? and the panel couldn't agree.

This year, Colorado lawmakers are debating a similar measure, but its sponsors concede they don't know whether the "driving while high" bill will pass.

In Washington state, the ballot measure on marijuana legalization includes a 5 nanogram THC limit.

The measure's backers say polling indicates such a driving limit could be crucial to winning public support for legalization.

"Voters were very concerned about impaired driving," said Alison Holcomb, campaign director for Washington's legalization measure.

Holcomb also pointed to a failed marijuana legalization proposal in California two years ago that did not include a driving THC limit.

The White House, which has a goal of reducing drugged driving by 10 percent in the next three years, wants states to set a blood-level standard upon which to base convictions, but has not said what that limit should be.

Administration officials insist marijuana should remain illegal, and Kerlikowske called it a "bogus argument" to say any legal level of THC in a driver is safe.

But several factors can skew THC blood tests, including age, gender, weight and frequency of marijuana use. Also, THC can remain in the system weeks after a user sobers up, leading to the anxiety shared by many in the 16 medical marijuana states: They could be at risk for a positive test at any time, whether they had recently used the drug or not.

A Colorado state forensic toxicologist testified recently that "5 nanograms is more than fair" to determine intoxication. But, for now the blood test proposals remain politically fraught, with supporters and opponents of marijuana legalization hinging support on the issue.

Huestis, of the government-funded drug abuse institute, says an easy-to-use roadside saliva test that can determine recent marijuana use ? as opposed to long-ago pot use ? is in final testing stages and will be ready for police use soon.

Researchers envision a day when marijuana tests are as common in police cars as Breathalyzers.

Until then, lawmakers will consider measures such as Colorado's marijuana DUI proposal, which marijuana activists say imperils drivers who frequently use the drug such as Chilton, the multiple sclerosis patient.

She says that since she began using pot she has started driving again and for the first time in five years has landed a job.

Chilton worries Colorado's proposal jeopardizes her newfound freedom.

Court blocks more parts of Alabama immigration law - Herrera ...

(Reuters) - An appeals court ordered the state of Alabama on Thursday to stop enforcing additional parts of its controversial new immigration law, pending review of a federal challenge to the measure that is considered the toughest in the nation.

The U.S. Court of Appeals for the 11th Circuit, based in Atlanta, issued a brief order expanding its initial injunction to include provisions that barred illegal immigrants from obtaining a driver's license and barred courts from enforcing contracts that involved illegal immigrants.

The appeals court in October blocked the state from enforcing another key provision of the law that required schools to check the immigration status of children on enrollment.

The provisions are blocked pending the outcome of the legal challenge by President Barack Obama's administration, which has argued that regulation of immigration should be handled by the federal government and not the states.

In its ruling in October, the appeals court also blocked a provision of the Alabama law, which was passed last year by large margins in both chambers of the Southern state's Republican-led Legislature, that made failing to carry documents proving legal residency status a misdemeanor crime.

That part of the law resulted in some embarrassing incidents when two foreign employees in Alabama's important auto industry were detained briefly for failing to produce proof of legal residency.

Businesses in the state, especially farmers, have also protested the law, saying it had caused widespread departures of Hispanic workers from the state, creating an employee shortage.

After the incidents, Alabama Governor Robert Bentley said he and the Republican leadership of the Legislature would review the new law, but not repeal it.

"I will continue to vigorously defend Alabama's immigration law in the courts," Alabama Attorney General Luther Strange said on Thursday. "I am hopeful that the Supreme Court's coming decision in the Arizona case will make clear that our law is constitutional."

Arizona in 2010 became the first state to pass a tough immigration law, parts of which were also blocked by the courts. The Supreme Court is expected to rule on the case, which could clarify whether states have a role in immigration policy.

Illegal immigration is a contentious issue in the United States, where an estimated 11.2 million illegal immigrants live.

In the Alabama case, the court on Thursday ruled the state could continue to authorize police to detain people suspected of being in the country illegally if they could not produce proper documentation when stopped for any reason.

Federal judges previously blocked key parts of other immigration laws passed in Georgia, Arizona, Utah and Indiana.

Mary Bauer, legal director for the Southern Poverty Law Center, said the court's action was noteworthy because there was no motion pending before it on the matter, so the ruling was of the three-member panel's own volition.

The SPLC is joined in opposition to the law by the American Civil Liberties Union, the ACLU of Alabama, the National Immigration Law Center, the Asian Law Caucus and the Asian American Justice Center.
